Dosing schedules of combination approaches

The dosing schedules for both tafasitamab and epcoritamab in combination with rituximab and lenalidomide are quite intensive initially, Poh says, but become less intensive over time. With tafasitamab, treatment is given intravenously once a week during the first three cycles, and then the frequency decreases to every other week through cycle 12. Lenalidomide is taken orally for 21 days of each 28-day cycle, while rituximab is given in the first five cycles. With epcoritamab, there is also an initial weekly step-up dosing schedule during that first cycle to try to mitigate the risk of cytokine release syndrome. The frequent visits during the early part of treatment can be beneficial, particularly for patients who have higher tumor burden, because they allow for monitoring of patients more closely for toxicity and early complications, she observes.
